Bcr-Abl kinase 표적이 소실한 imatinib 내성 만성 골수성 백혈병 세포에 대한 새로운 표적과 유효한 약물을 탐색하기 위하여, K562 세포로부터 3종의 imatinib 내성세포인 K562/Rl, K562/R2 및 K562/R3 세포를 분리하여 본 연구에 사용하였다. 이들 내성세포는 Bcr-Abl level이 현저히 감소되나, Bcl-2 및 Ku70/80의 발현은 증가되며, 또한 Bax, DNA-PKcs 및 BRCA1의 발현은 감소되는 특징을 가진다. 그러므로 이들 내성세포의 imatinib 내성기전은 Bcr-Abl 비의존적이며, 다른 cell survival 기전에 의한 것임을 밝혔다. 그리고 K562/R3 세포는 K562 세포보다 오히려 camptothecin (CPT) 및 radiation-induced apoptosis에 감수성을 나타내어, imatinib 내성세포는 DNA damaging agents에 과감수성의 특징을 가짐을 알 수 있었다. 한편, K562 세포에서 imatinib와 CPT를 병용하였을 경우, 이들 약물 단독 처리군에 비하여, Bax level 의 감소 및 PARP-1 및 DNA-PK의 cleavage가 증가되어, 이들 약물간의 병용효과가 있음을 알 수 있었다. 그러므로 본 연구는 CPT가 imatinib와의 병용효과를 나타낼 뿐만 아니라 Bcr-Abl kinase 표적이 소실된 imatinib 내성세포에 유효할 수 있음을 시사하였다.
