Tetrahydropapaveroline (THP)은 dopamine에서 Pictet-Spengler type의 축합 반응에 의해 생성된 tetrahydroisoquinolines (TIQs)로 L-DOPA 요법 파킨슨씨 질환 및 알코올중독 환자의 뇨 및 혈중에서 발견된다. 또한, MPTP와의 구조적 유사성으로 endogenous neurotoxin으로 간주되고 있다. 따라서, 본 연구에서는 THP가 PC12 세포와 신경분화된 PC12 세포중의 dopamine 생합성 및 L-DOPA 유도 세포독성에 미치는 영향에 대해 검토하였다.
THP는 PC12 세포에 24 시간 혹은 48 시간 전처치하였을 때 dopamine 함량을 농도 의존적으로 유의성 있게 감소시켰고, PC12 세포내의 TH 활성 또한 농도 의존적으로 저하시켰다. 반면, L-DOPA는 PC12 세포중의 dopamine 함량을 현저히 증가시켰지만, THP의 병용 처리에 의해 L-DOPA 유도 dopamine 함량 증가 작용을 저해하였다. 또한, 감소된 dopamine 함량은 항산화제인 NAC의 사용으로 회복되었다.
또한, THP를 L-DOPA와 병용 처리하였을 때 L-DOPA 유도 세포 독성에 미치는 영향에 대해서도 검토하였다. THP 10 μM 혹은 L-DOPA 20 μM과 같은 저농도에서는 MTT 분석에 의한 세포 생존율에 영향을 미치지 않았으며, TUNEL과 flow cytometry 분석에 의한 세포사도 유발되지 않았다. 그러나, THP 15 μM 및 L-DOPA 50 μM 이상의 고농도에서는 세포 독성을 유발하였다. 또한, THP를 L-DOPA와 병용 처리시엔 L-DOPA 유도 세포 독성을 현저히 증가시켰다. THP, L-DOPA 단독이나 병용 시 세포내 ROS의 생성은 현저히 증가시켰고, ATP 수준은 감소시켜 산화적 스트레스에 의한 세포사가 유발됨을 확인할 수 있었고, 항산화제인 NAC 처리에 의해 유발되는 세포사는 강력히 억제되었다.
THP의 신경 독성 작용을 확인하기 위해 NGF로 분화시킨 PC12 세포를 이용하여 세포내 dopamine 함량 및 L-DOPA 로 유도된 세포독성에 미치는 영향에 대해 검토하였다. 분화된 PC12 세포에서는 THP에 의해 15 μM 농도에서부터 유의성 있게 dopamine 함량을 감소시켰고, L-DOPA 와 병용 처리 시 L-DOPA 유도 dopamine 함량 증가 작용을 저해하였다. MTT 분석에 의한 세포 독성은 20 μM 농도에서부터 유의성 있게 나타내었고, THP를 L-DOPA 와 병용 처리시엔 L-DOPA 유도 세포 독성을 현저히 증가시켰다. PC12 세포에서 THP에 의해서 MAPK family 중 세포 생존과 적응에 중요한 ERK는 이른 시간에 활성화되었다가 감소하는 반면, 세포사와 관련된 JNK는 보다 오래 유지됨을 확인할 수 있었다. 또한, 부분적으로 세포 생존에 관여하는 p38은 ERK와 유사한 경향을 나타냈다. 분화된 PC12 세포에서도 비슷한 경향을 나타냈으나, 특히 ERK는 이른 시간에 활성화 되었다가 오래 유지되었다.
이상의 결과를 종합하면, THP는 PC12 세포와 분화된 PC12 세포에서 모두 dopamine 함량을 감소시키고, L-DOPA 로 유도된 dopamine 함량 증가를 억제하며, L-DOPA 세포독성을 증가시키나, 분화된 PC12 세포에서 그 독성이 완화됨을 확인할 수 있었다.
