Radiation method in head and neck cancer treatment is considered the most common cause to the severe problems of salivary gland dysfunction. It is believed that radiation causes the exhaustion of local tissue stem cells pool in salivary glands, thus resulting in its dysfunction. The present study succeeded in isolating human minor salivary gland stem cells which possess the following properties: 1) having high proliferation; 2) being capable of expressing multi-stemness markers as well as differentiating into many kinds of cell types. Human minor salivary gland stem cells in hypergravity and microgravity environment in a short period of time were also highlighted in the study. The results showed that the huMSGSC increased the proliferation in hypergravity and decreased proliferation in microgravity compared with control ground gravity. Furthermore, the stemness markers of huMSGSC also had higher expression in hypergravity group than control ground gravity group, but fluctuated in microgravity group. The cell-cell junction of huMSGSC, such as Zo-1, Zo-2 changed according to gravity change. These markers were upgraded in hypergravity group and downgraded in microgravity group, compared with control group. The huMSGSC also showed better capability of differentiation into hepato-cell like in hypergravity group. In conclusion, stem cells isolated from human minor salivary gland stem cells have the function of multi - potent stem cells. A short - term huMSGSC exposure in hypergravity is better for stemness expression and capability of differentiation. Cell-cell junction change under hyper gravity could play an important role in the results.
