In this study, I investigated the antioxidant and anti obese activity of herbal extract. Total phenol contents was 53.46±3.46 mg/g and total flavonoid contents was 4.42±0.17 mg/g in herbal extract. The antioxidant activities were evaluated through radical scavenging assays using 2,2-diphenyl-1-picrylhydrazyl(DPPH), 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonate) (ABTS), superoxide, hydroxyl radicals and reducing power. The DPPH radical and hydroxyl radical scavenging activities of herbal extract were 47.99±0.28%와 38.63±0.25% at 5 mg/mL, respectively. ABTS radical and superoxide radical scavenging activities were 47.58±0.31%와 55.90±3.07% at 1 mg/mL, respectively. The reducing power was 0.864±0.013, 1.192±0.001, 1.545±0.068 and 1.878±0.049 at concentrations of 2, 3, 4 and 5 mg/mL, respectively. Obesity was induced by feeding on high-fat diet(HFD) containing 40% kcal fat for 6 weeks, in C57BL/6J mice. The C57BL/6J mice were randomly divided into four groups; normal control group(NC), high-fat diet control(HFD), treatment group with fat down(HFD+fat down) and herbal mixture(HFD+herbal mixture) at 150 mg/kg body weight/day, per oral. Mice were administered with herbal mixture once a day for 8 weeks. At the end of treatment, we examined the effect of herbal mixture on body weight, plasma lipid, liver lipid and feces lipid. The administration of herbal mixture significantly decreased epididymal adipose tissue weights compared with HFD group. Significant decrease in the levels of triglyceride and total cholesterol in the serum were observed in the herbal mixture group compared with HFD group. HDL cholesterol level in the serum was significantly increased in herbal mixture group compared with HFD group. Total cholesterol and triglyceride level in the river were significantly lower in herbal mixture group than HFD group. In addition, total cholesterol and triglyceride level in the feces were significantly increase in herbal mixture group than HFD group. Histological analyses of fat tissues of mice treated herbal mixture decreased size of adipocytes compared to the HFD group.
These results suggest that herbal mixture extract have antioxidant and anti-obesity activities and can regulate serum and liver lipid profiles by reducing total cholesterol and triglyceride levels. In addition herbal mixture can promote the secretion of total cholesterol and triglyceride. In the future, herbal mixture can be useful ingredients in functional foods.
