스트레스는 어떤 물리적 또는 심리적 사건으로부터 생물체의 항상성이 위협받는 상태를 일컬으며 장기간의 스트레스는 중추신경계에 영향을 미친다. 이러한 영향은 CRF (corticostrophin-releasing factor), ACTH (adrenocortoctrophic hormone), CORT (cortisol, corticosterone) 등의 신경펩타이드와 신경물질의 변화를 초래하며 이는, 우울증과 외상 후 스트레스 등의 정신질환을 유발한다.
본 연구에서는 먼저 만성 스트레스 모델로 실험용 쥐에서 장기간으로 CORT를 투여함으로써 스트레스 모델을 확립하였다. 또한, 약물투여 후 트레드밀 운동을 시킴으로써 운동이 만성스트레스에 미치는 효과를 CRF, ACTH, BDNF의 분석을 통하여 알아보았다. 그 결과 장기간의 CORT 투여는 혈청의 ACTH의 농도를 감소시켰으며 또한, 뇌의 해마에서 CRF의 발현량이 급격히 감소하였고 BDNF의 발현량이 약간 증가하는 것을 보여 주었다. 장기간의 CORT 투여에 운동을 같이 복합처치 한 그룹에서는 감소된 ACTH와 CRF가 정상수준으로 올라왔으며 BDNF 발현량이 급격히 증가한 것으로 나타났다. 지금까지 많은 선행연구에서 스트레스는 HPA axis를 활성화시키고 BDNF를 감소시킨다고 보고되었다. 하지만 외상 후 스트레스 환자와 과도한 스트레스는 오히려 HPA 활성을 감소시키며 이러한 감소는 걱정과 불안 등 정신질환을 유발시킨다고 보고되었다. 따라서 우리가 확립한 만성스트레스 모델은 HPA axis의 활성을 감소시켰고 이로 인한 BDNF 발현량이 증가되었다고 사료되나 추후의 많은 연구가 필요하겠다. 본 연구결과 운동은 장기간의 CORT에 의한 HPA axis 억제를 활성화 시켰으며 해마의 CRF와 BDNF의 발현량을 증가시켰다.
따라서 본 연구를 통하여 도출된 결과들은 운동은 만성스트레스에 의해 발생된 질환들에 대해 긍정적인 효과를 미치며, 이는 장차 운동이 스트레스를 치료하는 목적으로 사용할 수 있는 가능성을 제시해 줄 것으로 사료된다.
